چکیده

Barron and coworkers (l--3) demonstrated nearly 30 years ago that the respiration of non-nucleated erythrocytes may be greatly stimulated by the addition of methylene blue to the buffer system. Explanations for this phenomenon have been given as increased oxidation of a degradation product of glucose (2), oxidation of lactate (4), increased reversible conversion of hemoglobin to methemoglobin (5), and conversion of lactic acid to pyruvate (6). None of these explanations, however, accounted for the magnitude of the increased respiration. From the studies presented here it is proposed that methylene blue activates a cyclic glucose oxidative pathway in mammalian red cells, and that this mechanism accounts for as much as 85 per cent of the oxygen consumed by human erythrocytes in the presence of methylene blue.

The effect of thiamine deficiency on the glucose oxidative pathway of rat erythrocytes.

Thiamine pyrophosphate has been demonstrated recently to be an essential cofactor for the transketolation of pentose to heptulose in the hexose monophosphate shunt in addition to its well known function in pyruvate utilization by animal tissues (l-4). The observation that the glucose oxidative pathway of mammalian erythrocytes may be studied in vitro in the absence of interfering oxidative phen...

Substances stimulating glucose catabolism by the oxidative reactions of the pentose phosphate pathway in human erythrocytes.

Mature human erythrocytes possess the enzymes necessary to catabolize glucose to CO2 via pentose phosphate pathway or to lactic acid via the Embden-Meyerhof pathway. These cells are deficient in certain enzymes of the tricarboxylic acid cycle. In the non-nucleated red cells, the oxidative reactions of pentose phosphate pathway appear to be the sole mechanism for glucose conversion to CO2 and fo...

Mechanisms of methylene blue stimulation of the hexose monophosphate shunt in erythrocytes.

The response of the hexose monophosphate shunt in erythrocytes was studied with the ionization chamber-electrometer apparatus to measure continuously 14CO2 derived from 14C-labeled substrates. The effect of methylene blue at high (0.1 mM) and low (1 muM) concentrations was evaluated under different gas mixtures; air, carbon monoxide, and 6% carbon monoxide in air. The latter gas mixture results...

Clinical Studies on Glucose Metabolism via the Pentose Phosphate Pathway in Human Erythrocytes.

Glucose metabolism via the pentose phosphate pathway was studied by measuring accelerated 02 uptake of human erythrocytes in the presence of methylene blue. Erythrocyte 02 uptake was independent of age in 67 normal patients. Erythrocyte 02 uptake was increased in pernicious anemia in relapse and in 5 of 6 jaundiced patients but was normal in pernicious anemia in remission and in 20 cases of mal...

Morphological and Morphometric Variations of Erythrocyte in Clarias Gariepinus, Bufo Bufo and Agama Agama Using Some Histological Stains

Introduction: Erythrocytes are highly specialized and the most abundant cell type in vertebrates’ blood. Their main function is the transportation of oxygen to tissues of the body via hemoglobin. Methods: The study was aimed at evaluating erythrocyte morphology in Clarias gariepinus (Cg), Bufo bufo (Bb), and Agama agama (Aa) using different stains; Also, morphometric analysis of erythrocyte w...
